usually, the Amp was gradually degraded, and nearly completely gone after 16
hrs.
and remember Amp is even unstable when sit at RT.


so the bacteria without the plasmid will grow faster than bacteria with it.
and u lose your Amp.


to get a high yield, grow up 5 ml first, then pour into a 1 L LB and grow o/
n.
