I have good experience with both, anti-sense RNA in situ can have very
high resolution, for example, if certain gene's expression is cell autonomous,
it can be shown as the only stained cell along many...
DNA FISH is harder to do, if you are observing whole mount tissue, you
probably will need a very good confocal or de-convolution microscope. I think
the quality of the probe is the most important factor...
Actually, I don't get your question, when you are doing in situ, should you
worry about the signal rather than the shape of the cell???
you should have provided more info...
