I think the priority of your question is to ask:

only the MMP promotor activity got affected?
if yes, this drug is specific to this (kind of) promotor
else this drug has general effect on
switch(case)
case 1: it affects transcription
case 2: it affects translation.
case 3: it affect protein exportation
case 4:............

qutitative RT pcr might answer some of your question, if you do Western and
find protein level didn't change, it then might be the protein exportation
problem...

actually, I have no idea, need to do more experiments to narrow the
possibilities.
