TUNEL labeling of tissue slides would tell you roughly how many apoptotic
cells are present on the slides. or you can try Hoechst staining for DNA
fragmentation. apoptotic cells are stained more bright for Hoechst, which is
thought due to change of chromatin structure. this is usually done with
propidium iodide staining, which stains dead cells bright. so apoptotic cell
is propidium iodide dim, and Hoechst bright. membrane structure change occurs
during apoptosis, and annexin V binds specifically to phosphoserine on
apoptotic cell surface, which could be used as a marker. flow cytometry is
combined with the techniques i mentioned above to score these cells by
different staining. search for more details. as well, i think it is fine if
you look at caspase activity, for example, caspase 12, which is suggested to
be specific to ER-stress-induced apoptosis, or caspase 3/9, etc, which are
linked to mitochondria-mediated apoptosis. look at proteolytic processing of
these proteins or direct caspase activity assay? not sure about the latter.
