1. Draft genome of sea squirt C. intestinalis 298:2157
Ciona diverged prior to the genome-scale expansions that
occurred in the vertebrate lineage, and this work tries
to understand the origins of chordate and vertebrate by
comparison of genome of Ciona with other invertebrates and
invertebrates. The genome has ~16,000 predicted protein-
encoding genes which show strong similarity to other
invertebrates, which about 1/6 of them have a vertebrate
counterpart involved in cell signaling and development.
2. Deletion of photoreceptive molecule melanopsin and
cricadian clock 298:2211, 2213
Understanding the molecular mechanism of circadian control
is facilitated with in vivo genetic manipulation of genes
that functions in the control. Melanopsin has been proposed
to be a photoreceptive molecule for mammalian circadian
system. Mice harboring deletion of melanopsin (OPN4) do not
exhibit overt defect in circadian activity, while they
display defective response to low light levels, and the
induction of early immediate gene is reduced. The data suggests
redundant signaling pathways exist. I wonder whether ppl
have addressed the genes that might respond to different
intensity of light (I guess different pathways should play
role, at least at the early phase, though they might converge
to a latter knot).
3. snRNP components in active spliceosome by MS analysis
298:2205
IP/Mass spec has been used to analyze the human 45S activated
spliceosome. (This method could be used to analyze protein-
protein interaction, components of transcripsome, etc).
