from ur second paragraph, the knockout mouse and the other antibody study,
it appears other members of the same cytokine family perform the
same role as yours in this pathway?
But u said previously that an antibody specifically targeting your
cytokine also blocked the effect? That antibody doesn't recognize other
cytokines within the same family?
It seems it would be pretty hard to justify these two results. If the
former one is the case, RNAi won't help.

: Hi, thank you guys a lot on your responses.
: We actually have done more. The molecule we thought play a role is a cytokine.
: Antibodies against other cytokines doen't block. If we give the cells this
: cytokine instead of the drug, it just did the same job, maybe even better.
: Something against the conclusion is that the knockout mouse still show same
: phenomina. But antibodies against cytokines within the same family can block
: such phenomina.
: Weired thing is I designed an experiment to block cytokine secretion but no
: difference. Elisa show the concentration of this cytokine has been reduced
: more than 70%. Do you guys think iRNA is still worth trying? I have never done
: iRNA before. How well could it block protein expression? I learned from
: commercial website that the efficiency for iRNA is 70~ 90%. Here is my
: concern. How could such little amount of cytokine play so important role.
: Especially because the phenomina it induced is a relatively one, it make me
: feel uncertain to believe it's mediated by a single cytokine instead of
: several cytokines.
: What do you guys think?
