It would be interesting if your parasite will trigger apoptosis. Usually
DNA breakdown is easy to detect 3-6 hours after mitochondrial membrane
potential loss, if your parasite does not have a direct effect on that.
However, there is a big concern. TMRE, derived from the classical potential
dye Rhodamine, will distribute along any potential, of course mitochondrial
membrane potential, but also other kinds of potential, such as plasma membrane
potential. Therefore, during your parasite infection, if there is a change of
plasma membrane potential, it's very likely that you will see a quick drop
of TMRE dye in the cells, although in this case, mitochondria will still have
a higher concentration of TMRE than the cytosol. The easy way to clarify
this is simply reloading your cells with TMRE, or if you are using realtime
confocal microscopy, you can supply some TMRE in the incubation buffer.
