You knockout mouse should be under the mixture genetic background of B6 and
129 at N1 generation while you WT might be the pure B6 or 129 background.

Because the genetic background could affect the phenotype depending which kind
traits you are studying, the reviewer need to make sure the difference you
observed is due to your KO gene but not the genetic difference.

TO get the pure B6 or 129 KO mice, you need backcross the N1 KO to either B6
or 129 at least five generations which could take at least one year.
