As ahche has pointed out, the change of cell size may be related with mTOR
pathway. It is possilbe that the k/n cells have impaired ability to sense
nutrients (aa, ATP, growth factors), and thus show lower rates of protein
synthesis and cell growth. If you want, u can check the phosphorylation of Akt
(Ser473 or Thr308), of S6K (Thr389), and of 4EPB1. If they (especially the
last two) show decreased level in your cells, it will indicate that the knock-
down can interfere that mTOR pathway (which has been believed to be a cell
size regulator).
Btw, how did you make such a high efficiency of knockdown? I have big problems
in transfection. In HeLa cells, lipo2000-mediated transfection of EGFP is
pretty ok, but any other plasmids (including EGFP-ubiquitin) have very low
efficiency, and the treatment of G418 leads to the loss of all transfected
cells. Tks
