Direct measurement for the binding of ATP to a ATPase is difficult to do.
Reasons are following. First, the affinity of ATP to a ATPase is often low,
around ~100 microM. This prevents studying the ATP binding using the
isothermal equilibrium binding measurements, either a filter-based assay or a
gel shift assay. Second, unlike GTPases that form a stable complex with a GTP,
ATPases usually hydrlyze ATP in a time frame that is faster than the time
required for most binding measurement. A nonhydrolysable ATP analog such as
AMPPNP and ATP gamma S, can partially overcome this hydrolysis problem.
However, the binding complex between a ATPase and these nonhydrolysable ATP
analogs is not stable, either. In addition, modified and labeled ATP analogs
are very expensive. Therefore, in order to measure the binding of ATP to a
