Yeast 2 hybrid can test protein--protein interaction( based on modular nature
of activator/ suppressors):
proteinA <--> proteinB+(AD) <-- Activation domain
+
(BD)<--DNA binding domain
-----------------------------------------------Reporter gene-----------DNA
Yeast 1 hybrid studies Protein--DNA interaction:
protein + (AD)
----------------------------Reporter gene---------------DNA
In 2 hybrid , fusion protein (pA+BD) binds to DNA(specific for BD binding),
but it does not have an activation domain(AD). So if pB can interact(bind)
with pA, then the whole complex can bind DNA and also contain an activation
domain which is fused to pB. Then Reporter gene expressed.
In 1 hybrid, Protein fused with an activation domain. If that protein can bind
to the seq. linked to the reporter gene, then the AD can activate the
transcription of the reporter.
I maight be wrong. But the above is what I remembered.
